THE DIABETES MILECULAR GENETICS SECTION OF THE PHOENIX EPIDEMOLOGY AND CLINICAL RESEARCH BRANCH AT THE NATIONAL INSTITUTE OF DIABETES AND DIGESTIVE AND KIDNEY DISEASES STUDIES STEM CELL DERIVED PANCREATIC ISLET ORGANOIDS COMPOSE OF MULTIPLE CELL. MANY EXPERIMENTS REQUIRE ISOLATION AND ANALYSIS OF SPECIFIC CELL POPULATIONS BECAUSE MIXED CELL POPULATIONS CAN PRODUCE UNCLEAR OR MISLEADING RESULTS. ALTHOUGH SINGLE-CELL SEQUENCING CAN IDENTIFY INDIVIDUAL CELL TYPES, IT IS COSTLY AND NOT PRACTICAL FOR ROUTING USES. AMORE EFFICIENT APPROACH IS TO SEPARATE CELLS BASED ON PROTEINS EXPRESSED ON THE CELL SURFACE. THE BD FACSMELODY CELL SORTGER WITH THREE LASERS AND NINE-COLOR CAPABILITY WILL ALLOW RESEARCHERS TO ANALYZE AND ISOLATE SPECIFIC CELL POPULATIONS FROM ORGANOIDS USING FLUORESCENT MARKERS. THIS CAPABILITY WITH SUPPORT DOWNSTREAM EXPERIMENTS FOCUSED ON CELL IDENTITY, MATURATION, AND RESPOSE TO EXPERIMENTAL CONDITIONS WHILE REDUCING RELIANCE ON MORE EXPENSIVE SINGLE-CELL SEQUENCING METHODS
